Not totally sure what you are asking here. You can set up 2 separate servers
and run one service from the existing server on one of the servers and run
another service from the existing server on the second machine.

Virtual server allows you to have a beefed up host server (RAM, disk space,
and CPU power) and you install other servers on that beefed up host and
divide the resources of the beefed up host between the installed guests.
Virtual server does not "turn an existing server" into anything. You install
one guest OS and set it up as a file server and install another guest OS and
set it up as a "separate application server".
FYI if the licensing has not changed, if you install the beefed up host with
Win 2k3 Enterprise you are entitled to install up to 4 guests operating
systems under the Enterprise licensing.

Virtual Server is an application program, not a magic potion.
How would you turn one physical server into two physical servers? When
you work out how to do that, you will be able to do much the same thing
using two virtual machines instead of the two physical ones.

It is hard enough to do P2V in the first place since the hardware
differs so much from the physical world inside a virtualized
environment. I know of exactly zero programs that will do what you
want on the fly.
Do yourself a break and create the virtual machines from scratch.
You can do one first and install the basic server operating system
including any security and other OS updates. Also install the VM
Additions.
Then you can clone this into two servers by copying the VHD file
(note: do not copy the vmc file) into a new folder and then create yet
another VS2005 guest while pointing it to the copied VHD disk. You
save yourself the OS installation time.
But you *must* run NewSID on the second server when you start it
because you cannot have two servers running with the same SID. This is
not an issue if you run the OS setup from scratch on the second guest
too (obviously with a fresh and empty VHD file in that case).
Then install any server role you want on these two servers.
